My 12v 180 amp alt puts out 12.8v @ 140 amps,will this step up volts to 14.2 (lithium)w/o changing amps?

This is not a buck boost or charger. It isolates batteries (two) such as start and windlass from another, typically larger, bank. It keeps the fully charged smaller batteries from contributing to the load on the larger battery, typically. You suffer a small voltage drop but otherwise it's transparent, and any change in incoming voltage would not be dependent on this unit. If your system is getting 14.2V, these will get 13.5V (0.7VDC drop).

Is it possible to hook two batteries in parrallel for charging from one post? So that 3 batteries are charging on two banks.

Yes, we actually have two batteries in parallel on each side with the alternator output connected to the center. Connected the way you are questioning Will provide the highest charge current to the battery with the lowest charge and this is what you want.

I don't see why not. Hook the charge controller output to the center alternator terminal, then each of the two banks to the other two posts. Make sure all three share the same physical ground and so long as you don't exceed the total amp rating of the isolator it should work fine.

Is this unit sealed? I want to mount it under the hood but I'm not sure if it's water resistant. It says "contaminant resistant."

Yes it’s a sealed unit. I installed mine under my air box on the front side of the passenger wheel well on a 2012 Sierra with no issue for a little over a year now
