Will this work on 2020 vehicles

No, can't update at all and no update since like 2019

Will this work on a 2006 dodge ram with Cummins 5.9 diesel?

It will work on anything 1996 and above as long as it has the OBD two plug which all 96 and above should have

Does the vehicle have to be running to get a reading or can the vehicle(key) be in "On (II)" position with this model?

The vehicle must be shutoof when you attach the computer and the turned to on but do NOT start the engine to run diagnostics. Once connected it will tell you on the diagnostics computer what to do.
